Under Edit\Preferences\Import, I tried to uncheck the box for "Show import dialog when a memory card is detected" but it doesn't "stick": when I close and open LR2 again, this option is checked again.

Go to Edit, select Preferences, Import tab and uncheck "show import dialog.....

I'm with Gary on this one, in Vista a pop up appears asking want you wanna do when it detects new stuff pluged in, you should be able to change the preferences there.
If always do this option, has been previously selected maybe the box don't pop up, so you will have to dig a bit deeper to change the settings.

I've got a back up programme that opens on the toolbar on boot up, think I can kill it, well not so far, it just sits there 'till I right click and close it.

If you feel 'daring', you could have a look at 'system management' \ System configuration. There is a tab there showing all Startup functions. Normally, you should find it there and you could disable it.
If you're on a PC, that is.
Yeah I have this problem, it's quite annoying because my LR database is on my external drive, so when I pop in my SD card before connecting the external, LR says it cannot find the LR database. When it is plugged in, windows just starts up the dialog box asking me what I want to do - it doesn't automatically open up LR. Show Import Dialog is definitely not checked.
Under startup I do see Adobe CS4 Service Manager - but if I disable this, what else, if anything, will I lose?
I don't want to uncheck "Use Autoplay for all media and devices" as that will effect everything.
bart it is likely that Explorer (assume you are using some flavour of Windows) has the file type registered to lightroom or the card reader's start option to LR.
