For me, the dedicated Pentax case that came with lens is handy for transporting from home to where I use it mostly to photograph birds, raptors mostly, is sufficient. I also have a Pelican case hard case with wheels and the pluck and pull insert fitted to the lens for serious travel. I need to take a photo with the lens in the Pelican case and post here, soon.

It can take the lens with the camera and grip attached, but I usually take only the lens in it for safety reasons (to avoid any lens mount wear).

Here is the Pentax DA 560 and the D_FA 150-450 in the Pelican 1615 Air. I have yet to pull the pluck foam to accommodate the 150-450 foot. There is plenty of room for a body or two and more lenses. Both lenses have Manfrotto QC plates installed, and both fit with the Pentax cases that came with the lenses with those mounted. The second shot showed the DA560 in it's Pentax case. It zips easily with the QC plate on. I do not leave the lenses in the Pelican as I'm not comfortable with them long term in the foam. I have considered using cotton cut from an old 't-shirt' to surround lenses in the pick and pluck foam when transporting in the weather sealed, roller Pelican case. The DA560 has a 3M vinyl camo product applied. I like it better than the neoprene style.

I use this NATX K9 Camera Bag - Elephant Grey - NATX Bags model. What I did was to ask the manufacturer owner (a wildlife photographer himself) to give two vertical separator. The 560 goes between those separators. And there are enough space to carry a few bodies+lenses, laptop also. And this case goes into most of the airline cabins without any issues.
