I am not sure what you are asking for. The light that comes on when focus lock is achieved? Or something else?

I need to know if it is possible in the K1II to "lock" the focus at a determinate point (subject) so that the focus stays the same even if the main subject changes.

Set it to back button focus. This disconnects the focus action from the shutter action. So you focus with the back button and it stays at that point no matter how many times you take a picture.

I set all of my cameras that way as soon as they come out of the box.

Assuming you are configured for other than continuous AF, holding half-press on the shutter should work as should holding down the AF button on the back.

You can also configure the AF button to "disconnect" the autofocus. That is, if you press and hold the AF button the camera will not re-focus when you press the shutter button. Sort of the opposite of what jatrax suggested. Either of those options will require you to do some menu diving, though. Steve's suggestion works right out of the box.

FWIW, I prefer to configure it the way jatrax describes. Takes some getting used to, but I think it gives you the best control.

Back button focus is probably the way to go. I used this with the K-5 and K-S2 but when I got my KP my fingers would not get used to the new button arrangement. I finally gave up and went back to using the shutter for a focus lock. If I want to lock the focus on a particular point after achieving focus I just switch the camera to manual focus. Usually, this is when making a panorama photo. I just have to remember to switch the camera back to autofocus afterward.

There is! Shutter button half press, with the camera set to AF-S, as several members have replied above.
Been that way since the very first Pentax autofocus film cameras and works the same on every digital Pentax camera.

But the OP asked for “one button operation” {in #8} - which to me means the shutter button when the camera is set “AF-S” mode as ‘Markii’ stated in #9.

If you are like me you will remember it later when you are reviewing your photos.
