I would actually prefer to have the buttons on the body like they are displayed on the grip in option A.
I could live with that as well.
It's very close to what I'm used to with the K-5 II and it works very well.

No real winner choice.

BIF photography, or anything that involves a subject that moves rapidly that needs tracking AF and the ability to start/stop AF at the drop of a hat.

Duh, stupid phone, I meant option A instead of B. Its easier to use the AF button on the side in Portrait mode. The camera is higher up and has a different weight distribution so your muscles will pick it up quite easily,
I'm with Robbiec here. At first I thought mimicking the body would be best, but testing my K-3 with DBG-5 it has the same layout as Option A. As it is in Option A, the thumb will rest easily on the AF-button in both modes, and the AE-L will be at the edge of the camera (not very nice to rest your thumb there). I use the rear AF button at all times (mostly doing portraits) so you would want your thumb to rest on the rear AF-button.

My thoughts exactly, comparatively speaking on the D4s the position of the AF-ON button was very well thought out ...the D3s was the problem.

Using back-button AF decouples taking a photo from focusing. This is great in dynamic situations, e.g., when you want to keep the same focus while taking several shots while re-framing all the time. With AF coupled to the shutter release button, one would always inadvertedly refocus.

With the AF button you can also just focus without having to worry about pressing a bit too much and inadvertedly taking a shot. In general, one can simultaneously take shots and focus but time each activity independently from the other.
