What woud be the best way for setting optimal focus?

I use something called a Bahtinov mask, which you can 3D print or order online for cheap in various forms. It works fantastic and has solved all my focusing issues.

Here's another shot from the same night, obviously the same subject, but stopped down to F5 instead of F4 and shot at 60 seconds instead of 40 seconds. I like it even better because I'm getting some starbursts on some of the highlights which makes for a nice look.

Wow! The amount of detail in the nebula is stunning! I need to buy a mask for the 300mm f4 or the DFA 150-450.

This is a 2-shot panorama with the SMC DA*16-50/2.8, stitched together in CS5. A good demonstration, I think, of the K-3iii's capability as a landscape camera.
