As a plug-in to Lightroom, Elements or Photoshop, and an extension to DxO Optics Pro, it will open anything they will, including DNG's.
Also the stand-alone version opens TIFF's and JPG's, so it's not too hard to integrate it into your DNG workflow.
Normally in using this kind of tool, I would open my original DNG first, do basic corrections (WB, exposure, clarity, noise reduction, sharpening, optical corrections etc) in my RAW processor, then fiddle about with filters and such on an exported version or virtual copy of the DNG original in TIFF or JPG format (depending on how much I cared about the quality of the image).
