Yes, the speed of the card matters, but you don't need a card that's faster than the camera can utilize.
I would think that a 30 to 45 MB/s is about the speed that the 645D can utilize given the number of years this camera has under its belt.
The 645D does not support UHS-I, so there is no need to spend extra for a UHS-I compatible card.
Note that the firmware must be upgraded to 1.01 or higher or else the 645D cannot work with SDXC cards. Given the size of the files you' maywant to use SDXC cards due to their higher capacity (64 GB and up).
Sandisk Extreme 45 MB/s, 32 GB or 64 GB cards would be a good choice.
