How does the 645 to K adapter work with the DA lens? Nobody has been able to show me how this would work.
I am afraid it probably wouldn't be very practical - since there is no electronic communication between the adapter and the camera, and since the 25mm has an electronic diaphragm, you wouldn't be able to change your aperture neither from the lens nor from the camera. This aside, you should be able to use MF without any problem.

The D-FA and DA lenses have a aperture lever that is mechanical, so if you have a 645-k adapter, does that control the manual aperture lever?

See here:

Pentax-K Adapter for 645 Reviews - Pentax K-mount Teleconverters and Adapters - Pentax Lens Reviews & Lens Database

"You will have to use stop down metering as the adapter neither has aperture contacts nor an aperture stop down lever". So no, you'll have to manually control your aperture - but at least you will be able to do so, whilst with the 25mm there is no way for you to manually control the electronic aperture of the lens.

Best,

Vieri
