1) You blow it off with a bulb blower.

2) If the above doesn't work, I use my wet cleaning kit. It's just some very pure alcohol and a lint free swab. Haven't trashed my Pentax yet and probably have done it about 10 times in two years.

Many methods, some good and some awful, are discussed below.
The Methods - Cleaning Digital Cameras - D-SLR Sensor Cleaning.

Hi, which camera do you have? Many current cameras have a dust removal mode that usually helps.
Otherwise use a dust blower gently. Don't touch anything with the tip.
Another option if you prefer or none of the above works, is to pay a shop to do it for you. It usually costs $40-50.

Sensor cleaning kit

Hi
I bought a Sensor cleaning kit at amazon
Amazon.com: Digital Survival KIT - Sensor Swab Type 2 (w/Eclipse): Electronics

It includes the "tools" you may need.I always thought that when you blow air into the sensor chamber you bring in new dust that will sit on the sensor after few days.
I have used it in my K100D and now on my K200D to remove dust that refuse to move even after 5-6 automatic sensor clean with the lans "looking" down.
Try to do it in a room with no wind and least dust (I do it in the bothroom).

In a pinch, a turkey baster does a decent fill-in as a bulb blower.
