The D300 will fare better if the test is biased towards overall responsiveness. It really does make the K20 look like a slug.
If your comparison must be done with everything set to default, then the D300 files will look punchier and sharper because the D300 defaults are higher than Pentax's.
If this is the way the test will be run, why not just read the DPReview tests? They've already done the work for you.
A fair comparison is when the user gets to optimize the output of the camera, and uses his lens at it's best aperture.
From reading your criteria, it looks like the D300 owner did some research and is setting the criteria to his camera's sweet spot. If he gets to do that, then you should be setting your camera up to it's sweet spot too.
