I guess you could look at the two methods this way:

The DOF method, you set the shutter speed you want, then adjust the aperture. So it's kinda like Shutter priority.

The AE-L method, you set the aperture and let the camera select the shutter speed. So it's Aperture priority.

I have tried both and I find the AE-L method to be the easiest. You can see the shutter speed that is selected in the viewfinder after you hit the AE-L button, and can adjust the aperture if you need a different shutter speed.

This topic has been very useful to me. My 50mm M 1.7 arrived today and I can't wait to play. I have one question; Do I have to press the AE-L button for every shot or only when I change the aperature? Thanks

That will depend on the lighting of the subject. If the lighting is constant, then you can take several pictures in a row. If the lighting changes (clouds moving in and out) then you should do the adjustment each time.

Also, you will have to press the AE-L button every time you shut the camera off.
