Tripod Collar

I got the 150-450 lens for Christmas which I am thoroughly enjoying. I'm having trouble removing the tripod mount for the first time from the lens. The locking knob becomes fairly tight after a few turns. Before I apply substantial force can the mount be removed and is it done by loosening this knob?
