If you are shooting outdoors anyway, I wouldn't be depending on flash. You probably don't have enough power anyway.
If there is a set of bleachers nearby (I'll use a ball diamond for this sort of shot), then you can pose the group on the bleachers. This combined with a lift will make for a nicer composition than having a mob of people on the ground all looking skywards.
You will want at least one helper, preferably two for a group this size.
If you use bleachers, you can sit the people down which will help keep your composition the way you want.
Make certain that you instruct your group that everyone should be able to see the camera with both eyes.
It's really hard to stay on top of group shots this big, the subjects themselves have a lot more to do with the success of a group shot this size than the photographer.
Personally, I don't think a DSLR has enough resolution for this type of shot, This is still medium format film territory.
